ATPase Pi is used to determine enzymatic ATPase activity by quantifying inorganic phosphate released during ATP hydrolysis. |
| Product description |
ATPase Pi is suited for ATPase activity measurements of myosins, S1, HMM and actomyosin. The assay operates in a moderate inorganic phosphate detection range of 50 µM to 1 mM, supporting robust measurement of ATPase activities where highly sensitive phosphate assays may be less suitable.
The assay is based on a modified Fiske–Subbarow method. Under reducing conditions, inorganic phosphate forms a blue phosphomolybdate complex. Organic phosphates remain unrecognized, making the assay compatible with phosphoproteins and nucleotides. |

Solution M: 1 × 100 ml molybdate solution; add 100 ml 2.6 N sulphuric acid.
Solution R: 1 × 100 ml reduction solution; add 100 ml ultrapure H2O.
Phosphate standard: 50 ml 1 mM KPO4, ready-to-use.
Standardized solutions in ultrapure H2O: 1 M imidazole pH 7.0, 3 M KCl, 0.1 M MgCl2, 0.1 M CaCl2, 0.1 M EDTA pH 7.0 and 0.1 M ATP pH 7.0. |
