Controlled source preparation
Skeletal muscle actin prepared from a single defined muscle type (musculus latissimus dorsi), to reduce biological variability.
Purity & concentration control
Purity assessed by scanning densitometry of Coomassie Blue G-250 stained SDS gels (15%). Protein concentration determined routinely by UV-VIS spectroscopy.
Functional polymerization checks
Polymerization performance of lyophilized actin is assessed batchwise using Ostwald viscometry and fluorometry with pyrene actin (10%).
Activity retention after lyophilization
Example benchmark: steady-state viscosity is no less than 95% of the actin prior to freeze-drying (batchwise QC criterion as stated).
Preservation approach
A dedicated cryoconservation method is used to preserve structural integrity and restore biological activity after storage and handling.
