Product description
|
Adenosine-5'-[(β,γ)-imido]triphosphate (AMP-PNP) is a non-hydrolyzable analogue of ATP. It is widely used in biochemical research to study ATPase activities, energy storage, and metabolism in vitro. Unlike ATP, AMP-PNP resists hydrolysis, making it a valuable tool for maintaining ATPases in a constitutively active state without the turnover typically associated with ATP hydrolysis. This property allows to dissect the mechanisms and dynamics of ATP-dependent processes more precisely.
Proteins such as actin and myosin, which exhibit functional ATPase activity, are often analyzed using AMP-PNP. In the case of myosin, AMP-PNP binds to the ATPase site and stabilizes the protein in a specific conformational state, facilitating the study of structural changes and interaction dynamics with actin. Similarly, for actin, AMP-PNP helps in examining the polymerization and depolymerization processes without the confounding effects of ATP hydrolysis.
Furthermore, AMP-PNP is utilized in various other studies, including molecular motor function, signal transduction pathways, and protein folding, where ATP binding and hydrolysis play crucial roles. Its application extends to the investigation of kinase activities, where AMP-PNP can act as a surrogate to understand phosphorylation events.
Overall, AMP-PNP is an indispensable reagent in molecular biology and biochemistry, enabling detailed exploration of ATP-dependent mechanisms and processes across a wide range of biological systems.
