| Target / specificity |
Clone 2G2 recognizes skeletal and non-muscle actin isoforms and has been used to detect cytoplasmic as well as nuclear actin forms. The antibody is conformation-sensitive and recognizes an epitope region that is accessible in profilin-complexed G-actin, but not exposed on the surface of filamentous F-actin. |

In immunofluorescence microscopy, methanol fixation supports detection of cytoplasmic actin, whereas paraformaldehyde fixation has been reported to reveal nuclear actin staining with clone 2G2. |

Clone 2G2 is particularly useful when actin conformation and fixation-dependent accessibility are relevant to the experimental question. Although classified as IgM, clone 2G2 is known to be very well detectable with secondary anti-mouse IgG antibodies and is also detected by anti-mouse IgM secondaries. This can be convenient if no dedicated anti-mouse IgM secondary antibody is available. As with all primary antibodies, optimal dilution and fixation conditions should be validated for the specific cell type and assay setup.

IgM. Clone 2G2 is known to be very well detectable with secondary anti-mouse IgG antibodies, although it is classified as an IgM antibody. It is also detected by secondary anti-mouse IgM antibodies. This is a practical advantage if anti-mouse IgM secondaries are not available in the laboratory, since additional anti-IgM secondary antibodies are usually not required for detection. |
