ATTO-labeled secondary antibodies combine strong fluorescence output with high photostability and reliable performance in fluorescence-based immunodetection. Donkey Anti-Mouse IgG (H+L) ATTO647N specifically detects mouse primary antibodies and is suitable for widefield, confocal and TIRF microscopy, flow cytometry, single-molecule detection and super-resolution imaging, including PALM, dSTORM and STED.
ATTO647N has an absorption maximum at 647 nm and a fluorescence maximum at 669 nm, corresponding to a Stokes shift of 22 nm. The conjugate is efficiently excited with 633 nm and 640 nm laser lines and is detected in the far-red spectral range. Its strong fluorescence output, high photostability and favorable far-red spectral characteristics support sensitive detection, repeated scanning and prolonged image acquisition in conventional, single-molecule and super-resolution fluorescence microscopy.
Donkey Anti-Mouse IgG (H+L) ATTO647N is antigen-specific; affinity purification removes essentially all donkey serum proteins, including immunoglobulins not specifically binding to mouse IgG. Following conjugation to ATTO647N, the antibody is further purified by size-exclusion chromatography to remove unconjugated dye and low-molecular-weight reaction components. The conjugate is supplied without preservatives or protein additives such as BSA.
