MonoMix is a defined G-actin buffer concentrate containing ATP, CaCl2 and DTT for standardized handling of monomeric actin. The buffer composition is selected to provide actin-compatible conditions during reconstitution, dilution and dialysis steps.
ATP and Ca2+ support the nucleotide- and divalent-cation conditions typically required for stable G-actin handling. DTT provides reducing conditions during short-term use after reconstitution. MonoMix is therefore suitable for preparing G-actin samples or ligand samples before controlled initiation of polymerization with an appropriate polymerization buffer. |

Each 1.0 ml 50× stock tube is sufficient to prepare 50 ml of 1× G-actin buffer. Total 1× buffer volume per unit: 2 × 50 ml = 100 ml. |

Final 1× buffer composition: 2 mM Tris-HCl pH 8.2, 0.4 mM ATP, 0.1 mM CaCl2 and 1 mM DTT. |
| Reconstitution |
Reconstitute each lyophilized tube with 1.0 ml ultrapure water to obtain the 50× stock. Dilute the stock 1:50 with ultrapure water to prepare the 1× working buffer. |
