Product description
Jasplakinolide is a macrocyclic peptide natural compound isolated from the marine sponge Jaspis johnstoni. It binds to actin filaments and inhibits their depolymerization by interaction at the interface of adjacent actin subunits within the filament.
Jasplakinolide is a potent inducer of actin polymerization in vitro because it promotes actin filament nucleation. It has high affinity for F-actin and lower affinity for actin monomers. Binding of Jasplakinolide strongly reduces the rate constant for dissociation of actin monomers from filament ends, thereby stabilizing filament structure and supporting workflows requiring persistent actin filaments.
Mechanism
Jasplakinolide stabilizes F-actin by binding to actin filaments, reducing monomer dissociation from filament ends and inhibiting actin filament depolymerization. It also promotes actin nucleation and can shift actin dynamics toward polymerized filamentous actin.
Functional notes
Jasplakinolide competes with phalloidin for actin binding. It is reported to inhibit the ATPase activity of F-actin and to stabilize filament structure by strongly reducing monomer dissociation, an effect associated with trapping of ADP within actin filaments.

Stock solution
To prepare a 500 µM stock solution, add 30 µl DMSO.
Alternatively, add 150 µl DMSO to prepare a 100 µM stock solution.
Application note
150 µl of a 100 µM Jasplakinolide solution stabilizes 2 mg actin at a 1:1 molar ratio.
Typical working concentration
Jasplakinolide is used for in vitro and in vivo assays at working concentrations from 50 nM to 5 µM. Incubation times range from 2 minutes to 24 hours. The optimal concentration and incubation time should be determined experimentally for each assay system.
Live-cell use
For live-cell assays, Jasplakinolide can be added to a variety of cell culture media, including complete media containing serum and antibiotics, provided that the final solvent concentration and compound concentration are compatible with the cell type and assay endpoint.
