Description
MotorMix II is a high ionic strenght buffer for soluble myosin II or to solubilize myosin II from the filamentous state.DTT and ATP are provided with the buffer to be freshly added before use.
The MotorMix II BufferKit consists of three components: MotorMix II, DTT and ATP. This allows preparation of two buffer variants used for myosin II.
The first variant is to supplement MotorMix II with 2mM DTT. This step is essential to prevent oxidation of myosin SH-groups. DTT is always added freshly to the MotorMix II, due its short half-life time.
The second variant is used to initiate the myosin ATPase by addition of e.g. 1mM ATP. ATP can be added either to the MotorMix II/DTT solution or mixed with the protein in MotorMix II/DTT.
