Actin monomer reservoir protein

Thymosin β4 protein for G-actin sequestration and actin pool control

A small, powerful actin-binding protein for creating defined pools of unpolymerized actin and controlling monomer availability in cytoskeleton assays.

Thymosin β4 binds monomeric actin in a 1:1 complex and keeps G-actin in an unpolymerized reserve state. By buffering the actin monomer pool, this tiny protein helps separate actin availability from immediate filament growth and gives experimental control over when actin can enter polymerization pathways.

Hypermol® Thymosin β4 is useful for reconstituted actin systems where spontaneous polymerization must be limited, monomer supply must be buffered, or the balance between sequestration and assembly must be studied. It complements Profilin workflows by providing a strong actin-monomer reservoir rather than promoting rapid ATP-actin delivery to elongating barbed ends.

Monomer sequestration
Forms 1:1 complexes with G-actin and limits spontaneous filament assembly.
Actin reservoir
Creates a defined pool of unpolymerized actin available after controlled release.
Assembly control
Helps tune the balance between monomer storage and filament formation.

For actin monomer buffering
Use Thymosin β4 to maintain G-actin in a soluble, unpolymerized state and to define how much actin is immediately available for filament growth.
For controlled actin assembly
Combine Thymosin β4 with actin, Profilin, Cofilin, capping proteins or nucleators to tune the transition from monomer storage to filament assembly.
For cytoskeletal homeostasis studies
Thymosin β4 is a useful tool for studying how cells store, release and redistribute actin monomers during cytoskeletal remodeling.
Strong experimental use cases
Create defined G-actin reservoir conditions in vitro.
Suppress spontaneous actin polymerization in monomer pools.
Compare Thymosin β4 sequestration with Profilin-mediated actin delivery.
Study controlled release of actin into assembly pathways.
