Toolkit description
The Actin-Toolkit SPR is designed for analysis of ligand interactions with filamentous cardiac actin by surface plasmon resonance (SPR). The assay format supports characterization of binding kinetics and dissociation constants for proteins, protein fragments and other actin-interacting ligands.
To reduce problems associated with long actin filaments in SPR fluidics, the toolkit uses short F-actin filaments anchored through AnkerproteinTM at the fast-growing barbed end. The anchored filaments protrude from the streptavidin-coated sensor surface and remain accessible to analytes introduced during the binding experiment.
The system can be used to investigate direct F-actin binding as well as the effects of pharmacological compounds, proteins or lipids on ligand–actin complexes. The toolkit contains native bovine cardiac actin, AnkerproteinTM, FAB-EP and GAB-EP SPR buffers and a dedicated handbook.
Streptavidin-coated sensor chips are required and are not included. Experimental conditions for the analyte or compound being investigated must be optimized individually.

Related products
For dilution of G-actin or exchange of ligand buffer into an actin-compatible buffer, MonoMix (Cat. #: 5100-0*) may be used. In the absence of nucleators, polymerization of G-actin can be initiated with PolyMix (Cat. #: 5000-0*).

Storage instructions
Store at −70 °C upon arrival. Kit components are stable for 6 months under these conditions. Solubilized G-actin should be kept on ice and used within 1 week. Avoid refreezing.
