MonoMix .01
G-actin buffer with ATP and 0.01 mM DTT; 100 ml (2 × 50 ml) supplied as lyophilized ready-to-reconstitute buffer.
Cat. #: 5120-01
Applications
| |
| Applications |
G-actin buffer for dilution, dialysis and handling of G-actin or G-actin-binding ligands under defined low-DTT, actin-compatible buffer conditions. |
| Recommended use |
MonoMix .01 is used when G-actin or ligands need to be diluted or equilibrated in a G-actin-compatible buffer with reduced reducing-agent concentration. It is suitable for workflows in which the standard MonoMix DTT concentration is not required or may interfere with downstream assay conditions. |

MonoMix .01 |
| Product description |
MonoMix .01 is a defined G-actin buffer containing ATP, CaCl2 and 0.01 mM DTT after reconstitution. Compared with standard MonoMix, MonoMix .01 provides the same general G-actin-compatible buffer system but with a substantially reduced DTT concentration.
ATP and Ca2+ support the nucleotide- and divalent-cation conditions typically required for stable G-actin handling. The low DTT concentration provides mild reducing conditions while minimizing the amount of reducing agent introduced into sensitive biochemical assays. MonoMix .01 is therefore suitable for preparing, diluting or dialyzing G-actin samples and G-actin-binding ligands before controlled initiation of polymerization with an appropriate polymerization buffer. |
| Overview |
Defined G-actin-compatible buffer system; supplied lyophilized for preparation of 1× working buffer; contains ATP, CaCl2 and low DTT; suitable for dilution and dialysis of actin and actin ligands under reduced-DTT conditions. |
| Related workflow |
MonoMix .01 is intended for G-actin-compatible dilution and equilibration steps when only a low concentration of DTT is desired. For initiation of actin polymerization in the absence of nucleators, PolyMix may be used as the corresponding polymerization buffer. |
Properties
| |
| Form |
Lyophilized, ready-to-use after reconstitution. |
| Quantity per unit |
2 × 50 ml after reconstitution. Each lyophilized vial is reconstituted with 50 ml ultrapure water to obtain final G-actin buffer.
Total G-actin buffer volume per unit: 1 × 50 ml = 100 ml. |
| Buffer |
Final 1× buffer composition after reconstitution: 2 mM Tris-HCl pH 8.2, 0.4 mM ATP, 0.1 mM CaCl2 and 0.01 mM DTT. |
| Reconstitution |
Reconstitute each lyophilized vial with 50 ml ultrapure water to obtain the 1× working buffer. Mix gently until completely dissolved. Keep the solubilized buffer on ice during use. |
| Purity |
Prepared from ultrapure reagents. |
| Storage instructions |
MonoMix .01 stored at −70 °C upon arrival will be stable in performance for at least 6 months from the date of purchase. Solubilized buffer is kept on ice and should be used within 1 week. Avoid refreezing. |
